Brian was born on 7 May 1964 to parents James Kilmeade and Marie Kilmeade in New York City, New York. He grew up with two brothers, Steve and Jim Kilmeade. He is of Irish and Italian descent from his family side.

Kilmeade with a height of 5 feet and 10 inches (1.78 meters) attended Massapequa High School. He then went to the C.W. Post Campus of Long Island University and graduated with a Bachelor of Arts at the age of 22.
